I am an online bookseller, and I want to add my site to the venues, like the many other (brick and mortar) bookshops.

LibraryThing local is not meant to be a directory of online booksellers

Management and members take a very hard line about online venues. Online venues must host bona fide, time-limited and interactive events of interest to book lovers. Selling books only is not sufficient.

My location on my home page (for local events) always comes up with one city. My location in my profile is set to another. How do I fix it?

There are two locations at work here. The location on your profile is public and can be seen by all. The location on your home page is private and can only be seen by you. The reason for this is that most people want to provide very accurate location information for the home module, especially in larger cities, so that events will be more accurate. But you don't want to provide house-level accuracy on your public profile for obvious reasons.

You can change your Home address by clicking on the pencil icon just above the Local section on your home page.
