Als Breck Coleman führt John Wayne hier einen Siedlertreck von Missouri nach Westen und sieht sich dabei von Schneestürmen bis hin zu Indianerüberfällen mit all dem konfrontiert, was ein solches Unternehmen zu einem lebensgefährlichen Wagnis machte. Aber auch der ungesühnte Mord an seinem Freund lässt Breck so lange nicht ruhen, bis er den Täter (Tyrone Power Sr.) im Schneesturm von Oregon endlich stellen und die Siedler an ihr Ziel führen kann. Inszeniert von Regielegende Raoul Walsh ("Die wilden Zwanziger", "Der Dieb von Bagdad") ist "Der große Treck nicht nur einer der ersten großen Tonfilm-Western überhaupt, er markiert gleichzeitig auch die erste Hauptrolle von Marion Michael Morrison, dem Walsh für den Film den Künstlernamen "John Wayne" verpasste. u.a. im Grand Canyon und weiteren Nationalparks entstanden, überzeugt der spektakuläre Frühwestern auch durch seinen Realismus in der Darstellung schier unmenschlicher Strapazen auf dem berühmten Oregon-Trail.
